The fixation protocol is intended for preserving tissues from autolysis and putrefaction.
Fixation may also increase the stability of protoplasm chemical components.
4% formaldehyde solution (usually named buffered formalin) is the most commonly used fixative in histopathology routine.
It is a non-coagulant fixative, it reacts with different tissue components hardening protein gels without separating water from proteins.
4% formaldehyde has a very low oxidation potential, it penetrates quite quickly into tissues while gradually hardening them.
Immerse completely specimen in a quantity of formaldehyde equivalent to 5 parts of the specimen volume.
Fixation time: at room temperature from 18 up to 24 hours, followed by a washing step of 1 hour with tap water.
